AP180 and Amphiphysin

To better understand endocytosis, we are investigating several other key components of the endocytic complex that act before dynamin.  AP180 is a scaffold protein that recruits the clathrin coat components to the vesicle.  Amphiphysin is another scaffold protein involved in recruitment of several endocytic proteins.  Both set the stage for the arrival of dynamin.  They also share the ability to co-ordinate a simultaneous interaction between proteins and cellular membranes.  Therefore they may be master regulators of endocytosis.  We have now identified all phosphorylation sites in both proteins, and are using this information to determine their specific function in order to map all the steps of endocytosis in the correct sequence.
